The effect of cycloheximide on the stability of the morphogenetic properties of chick limb-bud mesoderm☆
[摘要] Inthisstudydissociatedlatestage20(stage20+)chickleg-budmesodermcellsculturedincycloheximide(1μg/ml)weretestedtodeterminetheretentionoftheirabilitytoundergorecognizablelimbmorphogenesis,accordingtothetechniquedescribedbyZwilling(1964).Thecycloheximidetreatmentretardsthelossofthismorphogeneticpropertycomparedwiththenormallossthatoccursinculturesofuntreatedcells(FinchandZwilling,1971).Theexposuretocycloheximideextendstheperiodduringwhichthismorphogeneticpropertycanbedetectedforapproximately15–20hr.Incorporationexperimentsshowthatcycloheximideat1μg/mlrapidlyinhibitsthesynthesisofprotein.RNA,andDNAincellsexposedtotheantibiotic.Thisinhibitoryeffectiscompletelyreversibleinarelativelyshorttimeafterremovalofthecycloheximide.
