Possible roles for cell-to-substratum adhesion in neuronal morphogenesis☆
[摘要] Theroleofcell-to-substratumadhesionintheinitiation,elongation,andbranchingofaxonsfromembryonicsensoryneuronswasinvestigated.Cellsfromsensorygangliaof4–8-day-oldchickenembryoswereculturedonseveralsubstrata:includingcollagen;polyornithine-,polylysine-,andpolyglutamate-coatedsurfaces,andtissueculturedishes.Theair-blastermethodwasusedtomeasuregrowthcone-substratumadhesion.Growthconesadheremuchmorestronglytopolyornithine-orpolylysine-coatedsurfacesandtotheuppersurfacesofglialcellsthantotissuecultureplastic.Axons,too,adheretightlytothesesubstrata,andarecrooked,whereasontissuecultureplastic,axonsarenotadherentandarestraight.Thefractionofneuronsthatformaxonsandtheratesofaxonalelongationandbranchingaremarkedlyincreasedwhencellsareculturedonpolyornithine-coateddishesascomparedtotissueculturedishes.Thiscorrelationofstrongadhesionandenhancedneuronalmorphogenesissuggeststhatadhesiveinteractionsbetweenthegrowthconeandthemicroenvironmentinanembryoarecrucialpartsoftheinitiationandelongationofneuronalprocesses.Regulationofneuronalmorphogenesismaybeexpressedthroughthephysicochemicalpropertiesoftheinteractingcellsurfacesandextracellularenvironment.
