Maturation of rabbit reticulocytes: Degradation of specific reticulocyte proteins☆
[摘要] Asanalyzedbypolyacrylamide-gelelectrophoresis,rabbitreticulocytecytoplasmcontains,inadditiontoglobin,sevenpredominantpolypeptides.Theamountsoftheserangefrom0.1to1.2%ofglobin.Rabbiterythrocytescontainonlythreeofthesenonglobinpolypeptides.Thelossofthefourotherpolypeptidesiscorrelatedwithmaturationofthereticulocytestoerythrocytes.Wealsofractionatedreticulocytesaccordingtoagebyequilibriumcentrifugationthroughagradientofmetrizamideandshowedthatyoungerreticulocytescontainmuchmoreofthesefourpolypeptidesthandomorematurereticulocytes.Thefourpolypeptidesthatarelostduringdifferentiationarealsoverysensitivetoinvitrodestructionbychymotrypsinortrypsinunderconditionswhereglobinandthethreereticulocytenonglobinpeptidesthatremainduringreticulocytematurationarecompletelyresistant.Ourresultsareconsistentwiththenotionthatthedegradativerateofareticulocytecytoplasmicproteinisdeterminedbyitssusceptibilitytogeneralintracellularproteases.
