Localized ultraviolet laser microbeam irradiation of early Drosophila embryos: Fate maps based on location and frequency of adult defects☆
[摘要] Drosophilaembryoswerelocallyirradiatedwitha257-nmlasermicrobeamduringblastodermandgermbandstages.Dependingonstageandbeamdiameter(10–30μm),from0to45nucleiwereexposedtotheuvradiation.Thedosesused,5or10erg,didnoteliminatenucleiorcellsatonce,butupto50%oftheadultsurvivorsfromirradiatedeggscarrieddefectsinthethorax.Thesewerescoredwithreferencetotheimaginaldiscsfromwhichtheaffectedstructuresderive.Foreachthoracicdisca“targetcenterâ€wascalculatedastheweightedmeanvalueofallbeamlocationsaffectingtherespectiveadultderivatives.Thetargetcentersforthegermbandstagemapwithintherespectivegermbandsegments.Thepatternoftargetcentersfortheblastodermstageiscomparabletothethoracicregionofpublishedfatemaps,andthedistancesbetweenadjacentlegcenters(approximatelythreecelldiameters)agreewithrecentevidencebasedonmosaicflies.Wediscussthequestionwhetherthetargetcentersmarkthepositionoftherespectivediscprogenitorcellsatthestagesofirradiationandconcludethatthesepositionsarerenderedrathercorrectlyatleastwithreferencetothelongitudinaleggaxis.
