The identification of myogenic cells in regenerating skeletal muscle: I. Early anuran regenerates☆
[摘要] Muscleregenerationisusuallyconsideredtoinvolvetwoeasilyrecognizablemononuclearcellpopulations:(a)irregularlyshapedphagocyticmacrophages;and(b)fusiformmyogeniccellswhichresidebeneaththebasallaminaoftheinjuredmyofibers.Thepresentstudyfindsthatanuranmacrophagescanmimicearlymyogeniccellsbyadoptingafusiformshapeandasublaminarpositionduringtheinitialstagesofphagocyticinfiltration.ImplantsofmincedgastrocnemiusmusclefromadultRanapipiensandR.clamitanswereused.Macrophagecytologywasexaminedinnormallyregeneratingimplantsandimplantsmixedwithcarbontracerparticles.Implantsofnonlivingmuscle,lyophilizedorfrozenrepeatedlypriortoimplantation,werealsopreparedtostudytheprocessofmacrophageinvasionfreefromcontaminationbyanyendogenousmyogeniccells.Ultrastructuralexaminationofnormalimplantsfindsnoclearmorphologicalboundariesbywhichearlymyogeniccellsandmacrophagescanreliablybeseparated.Studiesofcarbon-markedimplantsdemonstratetheoccurrenceofnumerous,carbon-labeledfusiformsublaminarmacrophages.Fusiformmacrophagesarealsofoundinthemyofibersoftheimplantedmuscletissueskilledbypreviousfreezingorlyophilization.Theseobservationsindicatethatthecriteriaofcellshapeandlocationtraditionallyusedformyogeniccellidentificationareofdoubtfulvalidityandmustbereevaluated.
