Potential of isolated mouse inner cell masses to form trophectoderm derivatives in vivo☆
[摘要] Recentinvitroexperimentsonimmunosurgicallyisolatedmouseinnercellmasses(ICMs)havesuggestedthatsomeICMcellsmayretainthepotentialtoformtrophectodermafterinitialblastocystformation.Theseexperimentsreliedalmostsolelyoninvitromorphologyforidentificationoftrophectodermderivativesandprovidednoproofthattheputativetrophectodermcellswerecapableoffunctioninginutero.WepresentclearinvivoevidencethatatleastsomecellsinICMsisolatedfromearlyblastocystsdoretainthepotentialtoformpostimplantationtrophectodermderivatives.EarlyICMsoccasionallycontributedtotrophoblastfractionsinICM/morulaaggregationchimeras.Morestrikingly,whenaggregatedwitheachother,theseICMswerecapableofimplantingintheuterus,apropertyoftrophectodermcellsalone.Indeed,someaggregatesreconstitutedcompleteeggcylinders.However,ICMsisolatedfromlaterblastocystsrarelyproducedinvivotrophoblast,anditappearsthattheabilitytoformtrophectodermislostaroundthe16–19cellICMstage.Theseresultsarediscussedinrelationtochangingpatternsofgeneactivityinearlydevelopment.
