nhr-25, the Caenorhabditis elegans Ortholog of ftz-f1, Is Required for Epidermal and Somatic Gonad Development
[摘要] WehaveanalyzedtheexpressionandfunctionoftheCaenorhabditiselegansgenenhr-25,amemberofthewidelyconservedFTZ-F1familyofnuclearreceptors.Thegeneencodestwoproteinisoforms,onlyoneofwhichhasaDNAbindingdomain.nhr-25istranscribedduringembryonicandlarvaldevelopment.Anhr-25::GFPfusiongeneisexpressedintheepidermis,thedevelopingsomaticgonad,andasubsetofotherepithelialcells.RNA-mediatedinterferenceindicatesarequirementfornhr-25functionduringdevelopment:disruptionofnhr-25functionleadstoembryonicarrestduetofailureoftheepidermallymediatedprocessofembryoelongation.Animalsthatsurvivetohatchingarrestasmisshapenlarvaethatoccasionallyexhibitdefectsinsheddingmoltedcuticle.Inaddition,somaticgonaddevelopmentisdefectiveintheselarvae.TheseresultsfurtherestablishtheimportanceofFTZ-F1nuclearreceptorsinmoltinganddevelopmentalcontrolacrossevolutionarilydistantphyla.
