A cytochemical and ultrastructural analysis of in vitro maturation in amphibian oocytes☆
[摘要] Changesundergoneduringtheinvitromaturationofoocytesfrom8differentamphibianspecieshavebeenfollowedbycytochemicalmethods(Unnastaining,Feulgenreaction,PASreaction).CriticalstagesofmaturationinXenopuslaevishavebeenstudiedbyelectronmicroscopy.Attentionhasbeenmainlyfocusedontwotypesofstructures:1.1.Largebasophilicbodies,richinRNAandglycogen,whichformoraccumulateatthebasalendofthenuclearmembraneunderhormonalstimulation.Theyprobablycorrespondtopolysomalaggregatesandtheyseemdirectlyinvolvedintheruptureofthegerminalvesicle.Whenthebasalnuclearmembranebeginstoopen,thenuclearsapbecomesinvadedbyribosomesandglycogenandlateronbymitochondriaandsmallyolkplatelets.2.2.Feulgenpositivebodies,whichcorrespondtothenucleolarorganizers:theyappearatthebasalendofthenuclearmembranewhenthelatterbeginstobreakdown.Later,theymovetowardtheeggcortex,wheretheyultimatelyvanish.Theeffectsofactinomycin,puromycin,cycloheximide,andhydroxyureaontheformationofthesetwokindsofnucleicacid-containingbodieshavebeenstudied.Thesignificanceoftheseobservationsisdiscussed.
