Histochemical characterization of muscle differentiation☆
[摘要] Ahistochemicalstudyoffiberdifferentiationwasconductedonthelongissimusmuscleofpigsvaryinginagefrom1daytoapproximately24weeks.Thelongissimus,aswellasothermusclesofthepig,isfavorablefordevelopmentalstudiesbecauseofthecharacteristicarrangementofthethreefibertypeswithinthefascicles.Differentiationoffibertypeswasnotdetectableinthenewbornpig.Redfibersweredetectableby1week.Thedifferentiationofintermediateandwhitefibers,however,wasslower,butallthreefibertypeswereclearlydistinguishableby4-weeksofage.Throughoutdevelopmenttherewasagradualincreaseinareapercentcomposedofwhitefibersandadeclineinareapercentofbothredandintermediatefibers.Myosin-ATPaseandphosphorylasereactionsweremostsensitiveforearlydetectionoffibertypedifferentiationwhiletheNADH-TRreactionshowedagradualchange.ThedevelopingwhitefibersshowedasubstantialNADH-TRreactionforabout4weeks.Themeanfiberareaincreasedmorerapidlyinwhitefibersthanineitherredorintermediatefibers.Theunusualclumpdistributionofredfiberswasevidentatthesametimethatdifferentiationwasdetectable.
