Cellular interaction and chondrogenesis in vitro☆☆☆
[摘要] Theelaborationoforganizedtissuestructuresbydissociatedchondrocyteshasbeenanalyzedinvitro.Suchtissuereconstructioninvolvesnotonlysynthesisofmatrixcomponents,butalsotheorganizationofmatrixaroundthecellsinlacunae.Theexpressionofthistissuephenotypewasfavoredwhenaninitialinoculumofatleast106cellsperdishwasgrowninnutrientmediumCMRL-1066supplementedwith10%fetalcalfserum.AtthesamedensityinHam'sF-10orF-12(asmodifiedbyCoon)withthesameserumsupplement,thecellsproducedlessmatrix;andthematrixproducedwasnotorganizedaroundthecells.Growthonacollagensubstratedidnotalterthecells'responsetothevariousmedia.However,thedegreeofdifferentiationinF-10wasenhancedbysupplementingthemediumwithascorbicacid.Inclonalcultures(102cellsperdish)inCMRL-1066,well-differentiatedcoloniesarose.Thedevelopmentofthesecoloniesparalleledculturesinoculatedwith106cells.Culturesestablishedatintermediatecellpopulationdensitiesinthesamemedium,however,developedlargeareasofcellsnotassociatedwithorganizedmetachromaticmatrix.ItissuggestedthatinCMRL-1066cultures,chondrocytesnotonlyproliferate,secrete,andorganizematrix,butalsodegeneratelikehypertrophiedcartilagecellsinvivo.Becausethecellsinteractasapopulationbutdonotadapttoinvitroconditions,thisculturesystemmaybeusefulforstudyingtheprocessesinvolvedinthetransitionfromthecellulartothetissueleveloforganization.
