[摘要] Whengrowninasuitablemedium,chickchondrocytesretainthemorphologycharacteristicofcartilagetissueandsynthesizeamatrixwhosemainconstituentischondroitinsulfate.Ifchickembryoserumisaddedtotheculturemedium,thecellsattainafibroblastlikemorphologyandstopsynthesizingchondroitinsulfate.AstudyofthemetabolicfateofradioactiveglucosamineinwholecellshasrevealedablockatthelevelofUDP-NAG-pyrophosphorylasereactioninthefibroblastlikecultures.Thisblockcanexplainthecessationofchondroitinsulfateformation,sinceUDP-NAGisanimportantsubstrateinthepolymerizationofthisglycosaminoglycan.AconsiderablelevelofUDP-NAG-pyrophosphorylasewasdemonstratedinthehomogenatesofthefibroblasticcells.Variouspossibilitiesarediscussedconcerningthemechanismofthisblockanditsrelationtotheinhibitionofmatrixformation.
