Changing DNA polymerase activities during the development of the testis in the mouse☆
[摘要] DNA-dependentDNApolymeraseactivitiesfromthenucleusandcytosolhavebeenmeasuredduringdevelopmentofthetestisinthemouse.Analysisofsolubletesticularextractsofmicebetween5daysofageandsexualmaturity(38–42days)showedasix-tosevenfolddecreaseinthespecificactivityofthecytosolDNApolymerasefraction.Nofurtherdeclineinspecificactivitywasobservedbetweensexualmaturityand300daysofage.ControlexperimentshaveindicatedthatthisreductioninDNApolymeraseactivityisnoteasilyexplainedby:(i)adilutionofenzymeactivityduetoanincreaseinsolubleproteinsinthecytosol,(ii)adifferentialsolubilizationofenzymesduringdevelopment,or(iii)thepresenceofactivatorsorinhibitorsatspecificstagesofdevelopment.PartialpurificationofthetesticularDNApolymerasesbychromatographyonphosphocellulosesuggeststhatthedecreaseinspecificactivityofthecytosolisduetoadiminutionofactivityduringdevelopmentoftheDNApolymeraseα.Incontrasttothedeclineofspecificactivityobservedinthecytosol,thespecificactivityofthenuclearfractionincreasedtwo-tothreefoldduringdevelopment.
