A voltage clamp study of the sodium, calcium and chloride spikes of chick skeletal muscle cells grown in tissue culture
[摘要] Conventionalvoltageclamptechniqueswithmicroelectrodeswereappliedtochickmusclecellsgrownintissueculture.Thesimilaritiesanddifferencesinelectrophysiologicaldataobtainedfromnormalmyotubularmusclefibersandfromroundedmyosacs,producedbyincubationwithcolchicine,wereexamined.Undervoltageclampbothcellulartypesgeneratedthreedistinguishablevoltageandtime-dependentcurrentswhichcorresponded,respectively,totheNa+,Ca2+,andCl−spikesevokedunderconstantcurrentconditions.ThepresumedCa2+currentsweretoosmalltoallowquantitativecomparisons.Inmyosacs,butnotinmyotubes,therewasgoodcorrespondence,forboththeNa+andCl−systems,betweentheirspikethresholdsandpeakmembranepotentials,measuredunderconstantcurrentconditions,andtheircurrentthresholdsandreversalpotentials,measuredundervoltageclampconditions.Thiscorrespondenceisattributedtotheisopotentialityofthemyosacintracellularspaceandsuggeststhatmyosacsprovidemoreaccuratequantitativedatainvoltageclampstudiesthanmyotubes.
