Epithelial cell proliferation and migration in the developing rat gastric mucosa☆
[摘要] Toinvestigatecellproliferationindevelopinggastricantrumandfundus,proliferatinggastricepithelialcellswerelabeledinfetalratsbyintravenouslyinjectingmotherswith[3H]thymidine.Inaddition14-daypostnatal(dPN)ratsweregiven[3H]thymidineintraperitoneally.Tissuewasremoved1.5hrlater,andautoradiographswerepreparedtoidentifyproliferatingcells.Totalepitheliallabelingindices(L.I.)reachedapeakat20daysgestation(dG),coincidentwiththeappearanceofpit/glands,thendeclinedagainby22dG(gestationend).At18dG,labeledcellsweredistributedthroughoutalllevelsofthestratifiedepithelia.Between20and22dG,aspit/glanddevelopmentproceeded,labeledcellsbecameconcentratedintheglandbasesandwereonlyrarelyseenonthesurface(L.I.ofsurfacecells<1%at22dG).By14dPN,proliferatingcellswereentirelyabsentfromtheepithelialsurface.Approximately15%oftheendocrinecellswerelabeledat22dG,comparedtoonly2%at14dPN;zymogencellswerelabeled(∼6%)at14dPN;parietalcellsdidnotlabelatanyagestudied.Inaddition,cellmigrationtotheepithelialsurfacewasstudiedinratslabeledat22dG,14dPN,or28dPN,andkilled1–20dayslater.Migrationtimeswereslightlyshorterinthe28dPNgroupandwerelongerinfundusthanantruminallgroups.
