Changes in RNA and protein metabolism preceding onset of hemoglobin synthesis in cultured friend leukemia cells☆
[摘要] Wehavestudiedthehematopoieticprocesswhichisinducedbydimethylsulfoxide(Me2SO)intheOstertagFSD-1lineofFrienderythroleukemia(FL)cellsandhaveobservedseveralchangesthatprecedetheonsetofhemoglobinsynthesisat48hr.Althoughcellularviability,mitoticrate,anddeoxyribonucleicacidcontentareunaffectedbyourinductionprocedure,theinducedcellsbecomeprogressivelysmaller,andby96hrcontainonly55%asmuchribonucleicacidand60–70%asmuchproteinascontrolcells.Thedeclineinribonucleicacidcontentissignificantby24hrandaffects4Sandribosomalribonucleicacidsinanoncoordinatedmannerthroughoutthehematopoieticprocess.Furthermore,incorporationofradioactiveuridineintothe45Sprecursorofribosomalribonucleicacidisspecificallyinhibitedby1–2hrafterfirsttreatingFLcultureswith1%Me2SO.ThisearliestknowneffectofMe2SOonFLcellsisfollowedbyadeclineinsynthesisofprotein.Thebasicsequenceofmacromolecularandcellsizechangesaresimilartothosethatoccurduringnormalerythropoiesis.
