Study of differential collagen synthesis during development of the chick embryo by immunofluorescence: II. Localization of type I and type II collagen during long bone development☆
[摘要] ThetransitionoftypeIandtypeIIcollagensduringcartilageandbonedevelopmentinthechickembryowasstudiedbyimmunofluorescenceusingantibodiesagainsttypeIortypeIIcollagens.TypeIIcollagenwasfoundinallcartilaginousstructureswhichshowedmetachromaticstaining.TypeIcollagenappearedintheperichondriumofthetibiaatstage28andwasalsofoundinosteoid,periostealandenchondralboneafterdecalcification,periosteum,andtendons,ligaments,andcapsules.Usingtheimmunohistologicalmethoditwaspossibletoidentifyspecificcollagentypesinareasundergoingrapidproliferationandcollagentransition,suchasdiaphysealandepiphysealperichondrium,orinenchondralosteogenesis.DuringenchondralossificationtypeIcollagenisdepositedontotheerodedsurfaceofcartilage.Itpartiallydiffusesintothecartilagematrixforminga“hybrid”collagenmatrixwithtypeIIcollagen,whichisasiteforsubsequentossification.Duringappositionalgrowthofdiaphysealcartilageanddifferentiationofepiphysealperichondriumintoarticularcartilage,perichondralcellsswitchfromtypeItotypeIIcollagensynthesiswhendifferentiatingintochondroblasts.Inthetransitionzones,chondroblastsareimbeddedina“hybrid”matrixconsistingofamixtureoftypeIandtypeIIcollagens.
