Observations of the primordial germ cells in blood samples from the chick embryo
[摘要] Primoridalgermcells(PGCs)fromthecirculatingbloodin2-daychickembryoswereobservedbyhistochemicaltechniquesinsmearpreparationandbyphasecontrastmicroscopyinfreshsamples.Inthebloodsmear,PGCswerereadilydistinguishedfrombloodcellsbytheirlargesize(15–20μmindiameter)andbytheroundandlargenuclei(8–10μmindiameter)occupyingeccentricallythegreaterpartsofthecells.Histochemically,theyweredemonstratedtocontainabundantglycogen,alesseramountofyolkgranules(bythePASreaction),andalargeamountoflipiddroplets(bytheOilredOandSudanblackBstains)intheircytoplasm.Alkalinephosphataseactivitywasprovedslightlyintheircytoplasmbythesimultaneousdiazocouplingmethod.Inphasecontrastmicroscopy,livingPGCsinthebloodandfromtheprimitivegonadof3-dayembryosinpartsoftheexaminationswereobservedtochangetheirshapeinsomewhatamoeboidfashion,suggestingtheircapabilityforlocomotionasapossiblemechanismofthemigrationofPGCsinvivo.
