A reexamination of the leucine tRNAs and the leucyl-tRNA synthetase in developing Tenebrio molitor
[摘要] ThetranslationalcontrolmechanismpreviouslyproposedforthesynthesisofadultcuticularproteinsinTenebriomolitorisdependentupontheappearanceofamajor,novelleucinetRNAandachangeinleucyl-tRNAsynthetaseactivityjustpriortoadultemergence.Thepropertiesoftheleucyl-tRNAsynthetaseextractedfrompupaewerereexamined.Underoptimalaminoacylationconditions,nonewleucineisoacceptingtRNAswereobservedduringdevelopment.However,undersuboptimalconditions,adifferentialchargingoftheleucinetRNAspecieswasnoted.Thechromatographicprofilesofleucyl-tRNAsaminoacylatedinvivoinbothearlyandlatepupaewerefoundtobethesameandwereidenticaltotheprofilesobtainedbychargingtRNAsinvitro.PreviousevidenceforatranslationalcontrolsystemoperatinginTenebrioisdiscussedinrelationtotheseresults.
