Organ cultures of the embryonic rat parietal yolk sac: I. Morphologic and autoradiographic studies of the deposition of the collagen and noncollagen glycoprotein components of basement membrane☆
[摘要] Thesynthesis,deposition,andturnoverof[3H]proline-and[3H]glucosamine-labeledbasementmembranecomponentshavebeenstudiedinasystemusingorganculturesofembryonicratparietalyolksactissuesonanutrientagarsubstrate.Themorphologicandautoradiographicstudiesdescribedinthisreportwerecorrelatedwiththebiochemicalstudiesdescribedinthecompanionpaper(Minoretal.,Develop.Biol.48,1976).Thesestudiesshowedthat:(1)basementmembrane(BM)wastheonlyextracellularmatrixsynthesizedinthecultures,(2)10-to30-μmthicklayersofnewBMweredepositedduringa6-daycultureperiod,(3)itwasonlytheparietalendodermalcellsthatsynthesizedBM,(4)newBMwasdepositedonlyonthesurfaceofexistingBM,whichwasincontactwithendodermalcells,(5)theamountsofnewBMthataccumulatedwereincreasedbyatwicedailyfeedingscheduleanddecreasedbythepresenceoftrophoblast,(6)therewasanotabledifferenceintheredistributionoflabeledcomponentsduringa6-daychase,and(7)[3H]glucosaminetendedtolocalizeintheGolgicomplex,whereas[3H]prolinewasdistributedthroughoutthecytoplasmoftheendodermalcells.
