Use of preformed cell aggregates and layers to measure tissue-specific differences in intercellular adhesion☆
[摘要] Thebindingofaggregatesformedfromvarious7-daychickembryotissuestoculturedcelllayerswasanalyzed24hrfollowingtrypsindissociationofthetissues.Theproprotionofaggregatesbindingisindependentofthenumberofaggregatesadded,andchangeswithtimeover60mininamannerconsistentwithafirst-orderprocess.Theadhesiveparametermeasured,thepercentageofaggregatesbindingtocelllayersperunittime,variesslightlywithaggregatesizebutisnotdependentupontheprobabilityofcollisionoftheaggregatewiththelayer.Therateofbindingandtheeffectofmodifiersofbinding(temperature,inhibitorsofoxidativemetabolismandglutaraldehyde)aresubstantiallydifferentforneuralretinainteractionsthanforliverorheartinteractions,suggestingthatretinacellsmayformintercellularbondsviaamechanismdistinctfromthatofliverorheartcells.Therateofbindingbetweenliketissuetypesis,withoneexception,greaterthanbetweenunliketypes.Glutaraldehydetreatmentofonlyoneofthereactantsabolishesthisadhesivespecificity.Aggregatebindingprovidesameansofquantitativelyassessingintercellularadhesionwhichhastheadvantageofreducingtheeffectsoftrypsinizationonmeasurementsofadhesion,andthereforelendsitselftotheinvestigationofcellularconsequencesofadhesion.
