Autoradiographic study of protein and RNA formation during early development of Drosophila eggs
[摘要] PermeabilizedeggsofDrosophilamelanogasterwereincubatedintritiateduridine,valine,andphenylalanine.TheuptakeandincorporationintoTCA-insolublematerialweremeasuredbyscintillationcounting.Therewasverylittleincorporationofuridinebeforetheblastodermstage.Attheblastodermstage,theeggtookup2.4pmoles/hrofuridineandincorporated0.13pmolesintoRNA(assumingnodilutionofspecificactivityoftheprecursor).Theuptakeofaminoacidsvariedwiththeageoftheembryo;virgineggssynthesizedaboutasmuchproteinasfertilizedeggs.AutoradiographyofeggsincubatedinuridineshowedalackofRNAsynthesisinnucleiuntilthestartoftheblastodermformation.Thesmallamountofuridineincorporationbeforethisstagewasduetomitochondria.Incorporationofaminoacidswasuniforminthecytoplasmuntiltheblastoderm;therewasnoincorporationbyyolkgranules.Regionaldifferenceinlabelingappearedduringgastrulation.ThepolecellsdidnotformRNAduringtheblastodermstage,formationstartedduringgastrulation.Proteinlabelingofthepolecells,onthecontrary,wasverystrongintheblastodermandearlygastrula.Theseresultsindicatethattheexpressionofzygoticgenomebeforetheblastodermstageisunlikely.
