Isolation and characterization of mutations affecting UDPG pyrophosphorylase activity in Dictyostelium discoideum☆
[摘要] AprocedureforscreeninglargenumbersofclonesforanenzymeactivitywasusedtoisolatemutationswhichaffectUDPGpyrophosphorylaseactivity(EC2.7.7.9)inthecellularslimemoldDictyosteliumdiscoideum.FivestrainswererecoveredwhichhavelittleornoUDPGpyrophosphorylaseactivity.Tenotherstrainswerefoundwhichhavesignificantactivityinvivowhichisrapidlyinactivateduponcelllysis.ThesestrainshavepermittedustoevaluatetheroleofUDPGpyrophosphorylaseduringgrowthanddevelopment.Theenzymeaffectsthegrowthrateofthecellsbutisnotessentialforgrowth.However,duringdevelopmentthelackofenzymeactivityleadstocelldeathandlysis.StrainswhichlackUDPGpyrophosphorylaseaccomplishearlydevelopmentaleventsbutareunabletoculminate.However,certainbiochemicalandcytologicaldifferentiationsassociatedwithlatestageswereobserved.
