Trehalose accumulation in stalk and spore cells of Dictyostelium discoideum☆
[摘要] Single-celledmyxamoebaeundergodifferentiationintoeitherstalkcellsorsporecellsduringa24-hrperiodinDictyosteliumdiscoideum.Thisstudyemployedultramicrochemicaltechniquesandenzymaticcyclingtoassessthepresenceofcell-specificeventsinsporeandstalkcells.Freeze-driedsectionsofoneorganismwereassayedin0.1μlofreactionmixture.Thismethodwasusedtodeterminetheextentoflocalizationoftrehaloseinsporecellsandstalkcellsduringdevelopment.Trehalosewaslowintheearlystagesofdifferentiationtoabout20hrwhenthelevelstartedtoincrease.Indevelopingsporecells,thetrehaloselevelincreasedsixfoldduringthelast5hrofdevelopment.Likewise,theentirestalkcontainedtrehalosewhenthestalkwasfirstformed.Atmaturesorocarp,trehaloselevelswerethesameinsporesandtheapexofthestalk.Therewasadecreasinggradientoftrehalosedownthestalk.Thebottomone-fourthofthestalkwasdevoidofthisdisaccharide.Therefore,trehalosewasdegradedfromanareaofthestalkwhereitwaslocalizedearlierindevelopment.Theresultsofthisinvestigationnegatetheassumptionthattrehaloseisneverpresentinthestalk.Althoughtrehalosewasfoundinsporecells,prestalkcellsalsocontainedhightrehaloselevels.Thestalkcell-specifictrehalosewasnotretainedduringdifferentiation,however,butwasapparentlydegradedinthematurestalkcell.
