Inhibition of neurulation and interkinetic nuclear migration by concanavalin a in explanted early chick embryos☆
[摘要] EffectsofconcanavalinA(ConA)weretestedinchickembryosexplantedatstages4–7andculturedfor24hr.ConA(12–24μg/ml)blockedneuraltubeformationbyinhibitinginterkineticnuclearmigration,irrespectiveofthestageatexplanation.Somites,althoughlessnumerousthancontrols,werealmostnormalinappearance.Blastodermalexpansion,heartdevelopment,andbloodislandformationusuallywereunaffected.LightmicroscopicandautoradiographicstudiesshowedthattheapplicationofConA(16μg/mlorlower)causednoobviousvariationsincellmorphology,mitoticactivity,anduptakeof[3H]thymidineand[3H]uridine.ThegraftingexperimentsshowedthatConA(16μg/ml)stronglyinhibitedthedifferentiationofHensen'snodegrafts,butnottheirneuralinducingcapacity.Furthermore,theinhibitoryeffectofConAwasreversible,i.e.,embryosretainedtheabilitytorecoverfromsublethaleffectsofConAwhen,after4–5hroftreatment,subculturedonplainnutrientmedium.
