Cyclic AMP phosphodiesterase activity in fetal and adult muscle of the rhesus monkey☆
[摘要] TotalcyclicAMPphosphodiesteraseactivityofvoluntaryskeletalmuscleoftherhesusmonkeywashighestinthe100-dayfetalseries,decreasednearterm,andwaslowestintheadultseries.KineticdataindicatedtheexistenceoftwocyclicAMPphosphodiesteraseenzymesinboththefetalandadultmuscle.TheapparentKmvaluesforthehigh-affinityphosphodiesteraseweresimilarinthe100-dayfetalandadultskeletalmuscle,whereasthoseforthelow-affinityenzymeweretwofoldhigherinthefetalseries.TheVmaxofthehighKmenzymewastenfoldhigherinthefetalthanintheadultseriesandthelowKmVmaxwasfourfoldhigherinthefetalseries.BothcaffeineandtheophyllinewerecompetitiveinhibitorsofthelowKmphosphodiesteraseactivityandnoncompetitiveinhibitorsofthehighKmenzyme.NodifferencewasobservedinthesensitivityofthefetalandadultenzymepreparationstothemethylxanthinesortoRo20-1724.
