Retention of fully differentiated electrophysiological properties of chick embryonic heart cells in culture☆
[摘要] Manyculturedcellsinsphericalreaggregates(100–1000μmdiameter),preparedusingcellsisolatedfromembryonicchickventricles(16daysinovo),retaingreatsensitivitytotetrodotoxin(TTX;0.1μg/ml),rapidlyrisingactionpotentials(upto200V/sec),highrestingpotentials(upto−90mV),andtheylackautomaticity.TheNa+channelsgeneratingtheactionpotentialupstrokeinactivatetotallyatabout−50mV.Thechronaxie(hence,excitability),theratioofPNaPK,andtheintracellularK+concentrationareaboutthesameasinadultcells.FollowingblockadeoftheNa+channelswithTTX,norepinephrineproducesslowlyrisingovershootingresponses,indicatingthepresenceoffunctionalbeta-adrenergicreceptors.Thus,trypsin-dispersedmyocardialcellscanbemadetoretainhighlydifferentiatedmembranepropertiesinvitro.
