Hemoglobin synthesis in isolated erythroid colonies from the chick embryo☆
[摘要] Primaryculturesderivedfrommechanicallydissociateddefinitivestreakchickblastodermsweregrowninawarmairstreamonthestageofinvertedphasemicroscope,throughwhichinvitroerythroiddevelopmentcouldbeobserved.Proerythroidcellsdividethreeorfourtimesin48hrtogiverisetoerythroidcoloniesrangingfrom10to1000cells,dependingonthesizeoftheblastodermfragmentsfromwhichtheywerederived.Erythroidcelldevelopmentfollowsasimilarcourseinculturesgrowninacarbondioxideincubator.Coloniesconsistingofabout50cells,derivedfromblastodermfragmentscontaining5to10cells,wereisolatedandlabeledwith[3H]leucine,andtheirlabeledhemoglobinswereanalyzedbyisoelectricfocusing.Bothearlyhemoglobins(E,M,P,P′,andP″)andlatehemoglobins(AandD)aremadeincoloniesderivedfromsingleblastodermfragments.Theratiooflatetoearlyhemoglobinsisabout1.7inallcoloniesanalyzed.Theimplicationsofthisfindingfortheclonalmodeloferythroiddevelopmentarediscussed.
