Cell cycle changes during neural crest cell differentiation in vitro☆
[摘要] Chicktrunkneuraltubescontainingneuralcrestcellswereculturedinvitro.Celloutgrowthfromtheseneuraltubeexplantsconsistsprimarilyofasmallstellatecellpopulation.After3daysinculturethesmallstellatecellpopulationundergoesaremarkablechangeinmorphologythatischaracterizedbyamorerefractileappearanceinthephasecontrastmicroscope.Subsequenttothischangeinmorphology,pigmentgranulesbecomevisibleinthecytoplasmafter4daysinculture.After6daysinculture,virtuallyallofthesmallstellatecellsarepigmented.Thecellcycleparametersofthesmallstellatecellpopulationare:S=4.4±1.2hr(SD).G2=1.5±1.0hr(SD).M=1.7±0.6hr(SD).andGl=3.8±1.0hr(SD).Continuouslabelexperimentsdemonstratethat(G1+G2+M)increasesfrom7hrinDay4cells,asyetunpigmented,to12hrinDay5cellsthathavebecomepigmented.ThischangeisconsistentwithanincreaseinG1and/orG2thatiscloselycorrelatedwiththeappearanceofpigmentgranules.Itisofinterestthatthiscellcyclechangeiscorrelatedwitharatherlateeventinthedevelopmentalprogramoftheseneuralcrestcellsratherthanwiththeearliermorphologicalchangeobservedafter3daysinculture.
