Polysome-dependent synthesis of embryonic proteins of Artemia salina during cell differentiation and analysis of heme-containing protein☆
[摘要] ThepolysomesisolatedfromthedifferentdevelopmentalstagesofArtemiasalinahavebeentranslatedinahomologousS-30extractorpH5enzymefractionaswellasinaheterologous(wheatgerm)pH5enzymefraction.Thespecifictemplateactivityofthepolysomesincreaseslinearlyuptoastagewhichisequivalenttoabouttwo-thirdsofthewholepreemergenceperiodandthereafterremainsratherconstantuntilthenaupliusstage.Thespecifictemplateactivityofthepolysomesappearstobedependentonasourceofinvitrosystems,butnottobedependentonthesizeofthepolysomesused.Moreover,theefficiencyofpolypeptidechainreleasefromthepolysomesinvitroalsoseemsdependentonthedevelopmentalstagesoftheembryos,butinthiscaseitisnotinfluencedbytheinvitrosystememployed.Analysisoftheproteinssynthesizedinvitrobythepolyacrylamidegelelectrophoresishasrevealedthatthespeciesandtheirrelativeamountsoftheproteinsareratherspecificforeachdevelopmentalstageanalyzed.Heme-containingproteinsfromthenaupliihavebeenpartiallypurifiedandanalyzed.Thisheme-proteincomplexcontainstwodistinctpolypeptidechains,MW>100,000andMW=50,000–60,000daltons,respectively,asthemajorspeciesasjudgedbySDS-polyacrylamidegelelectrophoresis.Duringpreemergencedevelopment,noheme-proteincomplexeshavebeendetected,butafterhatchingoftheembryos,theyhavebeendetected.
