The relation between villus formation and the pattern of extracellular fibers as seen by scanning microscopy☆
[摘要] Duodenalmorphogenesisinthechickembryohasbeenstudiedtoseeifchangesinorganizationofextracellularfibersareassociatedwithvillusformation.Threemajorprocessesoccurduringmorphogenesis;longitudinalridgesform,thentheseridgesbecomezigzaginshape,andfinallyadoublerowofvilliformfromeachzigzagridge.Tissuesofdifferentdevelopmentalstageswereprogressivelytrypsinizedtoremovecellularmaterialandwerepreparedforscanningmicroscopytoshowtheorganizationofextracellularfibers.Resultsshowthatfibersystemsofincreasingcomplexityformasthedudoenumdevelops,andsuggestthatsomecellulareventssuchasinitialridgeformationprecedethesechanges.Tissueswithlongitudinalridgescontainonlyrandomlyorganizedfibers.Intissueswithzigzagridges,orientedfibersappearalongtheridgesandsomeextendlaterallyfromflexuresofeachzigzagridge,butinterridgefibersarestillrandomlyorganized.Whenvilliform,fibersinthebodyofthevillusarerandombutfibersatthebaseofvilliandbetweenvilliarehighlyoriented.Transmissionmicroscopyandcollagenasetreatmentofpartiallytrypsinizedtissueindicatethatmost,ifnotallextracellularfibersviewedbyscanningmicroscopyarecollagenous.Therefore,sincecollagenfiberorganizationissocloselyrelatedtomorphogeneticchanges,wepresumeitplaysanimportantrole.
