Number of mitochondria and some properties of mitochondrial DNA in the mouse egg☆
[摘要] Thenumberofmitochondriastoredinthe1-cellmouseeggwasdeterminedbyelectronmicroscopicmorphometry.Inthefoureggsexamined,theaveragemitochondrialnumberobtainedpereggwas92,500±7000SD.ThetotalDNAfrom6600unfertilizedmouseeggswasisolatedbybuoyantdensitycentrifugationinanethidiumbromide-CsClgradient.AfluorometricassayperformedonasampleofthepurifiedDNAgaveavalueofabout8pgfortheDNAcontentoftheunfertilizedmouseegg(withoutthefirstpolarbody);itwasestimatedthataboutone-thirdofthisamount,2–3pg,representedmitochondrialDNA(mtDNA).Electronmicroscopicvisualizationrevealedthat11%ofthemtDNApopulationconsistedofcatenatedforms.About30%ofthemtDNAcirclescontainedaDloopand70%werecleanduplexcircles.NolargerreplicativeintermediatesofmtDNAwerefound,suggestinganabsenceofmtDNAreplicationintheunfertilizedmouseegg.
