Cell interactions and the regulation of cholinergic enzymes during neural differentiation in vitro☆
[摘要] Seven-day-oldchickembryoneuralretina(NR),telencephalon(T),opticlobe(OL),andrembencephalon(Ro)weredissociated,andtheresultingcellsuspensionswereallowedtoreaggregateinvitroduring3dayseitherindependentlyorindifferentbinarycombinations.Interactionscouldbedetectedbythecomparisonoftheactivityoftheenzymesofthecholinergicsystem,cholineacetyltransferase(CAT)andacetylcholinesterase(ACE),in“pureâ€and“combinedâ€aggregates.Theresultsclearlyshowthattheactivityofbothenzymesinembryonicneuralcellscanbemodifiedselectivelybyinteractionsbetweendifferentcellpopulations.Thus,combinedNR-OLaggregatesshowanincreaseinCATwithoutchangesinACE,NR-TanincreaseinCATandadecreaseinACE,T-RoadecreaseinbothCATandACE,andOL-Tnochangesatall.ExperimentsinwhichNRandOLcellswerecombinedindifferentproportionsindicatethattheinteractionsrequirethepresenceofdefinednumbersofcellsfromeachkind.Isochronousandheterochronouscombinationsof7-and10-day-oldNRandOLcellsshowthattheinteractivecapacitiesofthecellschangewithdevelopment.
