Physiological and structural properties of colchicine-treated chick skeletal muscle cells grown in tissue culture
[摘要] Chickmusclecellsgrownintissue-culturemediumcontainingcolchicinedevelopedintoroundedcellscalled“myosacs.â€Electronmicrographsofmyosacsweresimilartountreatedmyofibersexceptthatmicrotubuleswereabsentandthecontractilematerialwasdisorganized.Mostoftheelectrophysiologicalcharacteristicsofmyosacsweresimilartothoseofuntreatedmyofibers.Thus,bothcellulartypeshadsimilarrestingpotentials,nonlinearcurrentvoltagecurves,threetypesofactionpotentials(Na+,Ca2+,andCl−spikes),andacetylcholinesensitivitywith“hotspots.â€Bothdemonstratedcontractionwithelectricalorchemical(acetylcholine,caffeine)stimuli.Theonesignificantdifferencewasthatmyosacs,unlikemyofibers,werealwaysisopotentialthroughouttheirintracellularspace.
