Transcriptional diversity in myogenesis☆
[摘要] WehavemeasuredthechangeinRNAtranscriptdiversityduringinvivomyogenesisinthechicklimbbymeansofsaturationhybridizationofwholecellRNAtononrepetitiveDNA.Wefindthattranscriptdiversitydecreasesasproliferatingmyoblastsfusetoformmultinucleatemyotubes.Specifically,RNAfromlimbmesoblastsat,orabout,thetimeofphenotypiccommitment(stage24,Day4.5)hybridizestotwiceasmuchnonrepetitiveDNAasdoesRNAfromfullyfusedmuscle(Day18ofdevelopmentthroughadulthood).Theleveloftranscriptdiversityfoundatstage24remainshighthroughouttheproliferativephaseofmyogenesis(throughDay12)anddiminishesduringthefusionphase(Days10–18).AdditivehybridizationexperimentsindicatethattheRNApresentduringfusionishomologoustoRNApresentinproliferatingmyoblasts,suggestingthatthechangeintranscriptdiversitydoesnotincludelargescaletranscriptionofpreviouslyuntranscribedsequencesduringthisinvivodevelopmentaltransition.
