Acceleration of amphibian embryonic melanophore development by melanophore-stimulating hormone, N6,O2-dibutyryl adenosine 3′,5′-monophosphate and theophylline☆
[摘要] Stage14(GallienandDurocher,1957)Pleurodeleswaltliiembryosweretreatedwithα-orβ-melanophore-stimulatinghormone(MSH),ACTH,dbc-AMP,c-AMPplustheophylline,theophylline,5′-AMP,or2′,3′-AMP.Thedevelopmentofmelanophoreswasaccelerated(appearingtwostagesearlierthanincontrolembryos)byα-orβ-MSH,ACTH,dbc-AMP,c-AMPplustheophylline,ortheophyllinealone.Bythetimeembryosdevelopedtostage28,thecontrolandtreatedembryoswereindistinguishableinthenumber,distribution,andgeneralmorphologyoftheirmelanophores,suggestingtheseagentsdonotinducemelanophoreformationormitosis.CyclicAMPalone,5′-AMPor2′,3′-AMPwereineffective.Acceleratedcytodifferentiationapparentlyrequirespreinductionbytheinvaginatingchordo-mesodermduringstages13and14aspresumptiveneuralplateexplantsculturedbeforechordo-mesoderminduction(stage8)failedtoproducemelanophoreswithorwithoutα-MSHordbc-AMP.Explantsfromstage14embryostreatedwithα-MSHordbc-AMPdevelopedthesamenumberofmelanophoresattheapproximatetimethattreatedwholeembryosdevelopedmelanophores.WholeembryoexperimentsinvolvingthefasterdevelopingembryosofXenopuslaevisweresimilartothosedescribedforP.waltlii.
