Sequential gene activation by ecdysone in polytene chromosomes of Drosophila melanogaster,: III. Consequences of ecdysone withdrawal☆
[摘要] EcdysoneinducesasequenceofchangesinpuffingactivityofthepolytenesalivaryglandchromosomesofDrosophilamelanogaster.Inthecontinuouspresenceofecdysonethissequencetakesabout12hrtocomplete.Salivaryglandswereincubatedinthepresenceofecdysoneforvarioustimesandthentransferredtohormone-freemediumtostudytheeffectsofthehormone'sremovalonthesequenceofpuffingactivities.Theearlyecdysone-inducedpuffsimmediatelyregressonremovalofhormone.Theycanbefullyreinducedifexposedtohormoneagain,butonlyiftheinitialperiodofexposuretothehormonewas2hrorless;withlongerperiodsofinitialexposuretheearlypuffsbecomeprogressivelyrefractorytoreinduction.Thisprocessisinhibitedbycycloheximide.Agroupofthelatepuffsmaybeinducedprematurelybyremovalofthehormone—theextentoftheirprematureinductioniscorrelatedwiththesizeoftheearlypuffsduringtheperiodofecdysoneexposure.Ecdysoneinhibitstheprematureinductionoftheselatepuffs.
