Derivation of the membrane comprising the male pronuclear envelope in inseminated sea urchin eggs☆
[摘要] Investigationswereconductedinanefforttodeterminetheoriginofthemembranecomprisingthemalepronuclearenvelopeofinseminatedseaurchineggs.Theeventsoffertilizationinzygotestreatedwith200μg/mlofpuromycinarenotimpairedeventhoughincorporationof[3H]leucineisinhibitedupto80%whencomparedtocontrolspecimens.Developingmalepronucleiinzygotestreatedwithpuromycinformnuclearenvelopesstructurallysimilartoandwithinthesameperiodascontrols.Inpuromycin-treatedanduntreatedzygotesmorphologicallyrecognizableportionsofthespermnuclearenvelopeareincorporatedintothestructureofthemalepronuclearenvelope.Pronucleardevelopmentwasalsoexaminedininseminatedovawheremostoftheendoplasmicreticulum(ER)wasconfinedtoaspecificareaofthezygote.Eggswerecentrifugedinordertostratifytheirorganellesintospecificlayers(stratifiedeggs);withfurthercentrifugationstratifiedeggsarebisectedtoformnucleate(richinER)andnonnucleatehalves(containinglittleER).ObservationsofinseminatedstratifiedeggsandnucleateandnonnucleatehalvesdemonstrateaninverserelationbetweentheamountofERpresentinthevicinityofareorganizingspermnucleusandthetimeittakestoformthemalepronuclearenvelope.Computationofthemaximumquantityofmembraneinthemalepronucleusthatmaybederivedfromthespermnuclearenvelopeisapproximately15%.Theseinvestigationssuggestthatamajorportionofthemalepronuclearenvelopeisderivedfromendoplasmicreticulumwithintheeggandonlyasmallportion(upto15%)originatesfromthespermnuclearenvelope.
