Undersulfated chondroitin sulfate in the cartilage matrix of brachymorphic mice
[摘要] Homozygousbrachymorphic(bmbm)micehaveadisproportionatelyshortstature,similartohumanachondroplasia.Wepreviouslyshowedthateachzoneofgrowthinyoungbmbmepiphysealcartilagesissmallerthannormalandthattheextracellularmatrixappearstocontainnormalcollagenfibrils,butsmallerandreducednumbersofproteoglycanmatrixgranules.Ourstudiesreportedhereindicatethatmutant,likenormalcartilage,synthesizestypeIIcollagenandcontainsnormalquantitiesofglycosaminoglycansasjudgedbyuronicacidcontent.However,theglycosaminoglycansfromthemutantdifferfromthenormalintheirchromatographicandelectrophoreticproperties.Furtherstudiesestablishedthatglycosaminoglycansfromcartilagesofbrachymorphicanimalswereundersulfated.WhereaschondroitinasedigestsofglycosaminoglycansfromcartilageofnormalC57Bl6J5-day-oldmicecontainedpredominantlydisaccharidessulfatedinthe4-position,thatofthemutantcontainedappreciableunsulfateddisaccharidesaswell.
