Inhibition of palatal epithelial cell death by altered protein synthesis
[摘要] Programmedcelldeathoccursinadefinedregionofthesecondarypalatalepithelium(medialedge)duringitsdevelopmentinvivoorinvitro.Thepurposeofthisstudywastoexaminetheeffectsofvariousinhibitorsofmacromolecularsynthesis,particularlyglycoproteinsynthesis,onthedeathofpalatalmedial-edgeepithelialcellsinvitro.RatpalatalshelvesexplantedongestationalDay15andculturedsinglyshowedmedial-edgecelldegenerationafter48hr,whereasinthepresenceof6-diazo-5-oxo-l-norleucine(DON),2-deoxyglucose(DOG),orcycloheximide,celldeathwasprevented.ThiswasincontrasttothelackofinhibitionofcelldeathinthepresenceofactionomycinDorcytosinearabinoside.SelectivelyaddingmetaboliteswhichbypasstheeffectofDONonvarioussyntheticpathwaysdemonstratedthattheinhibitionofcelldeathbyDONwasmostlikelyduetoablockinglucosamineformation.ThesynthesisandactivityofvariouslysosomalenzymeswerenotaffectedbyDON,whereasamarkeddecreasewasobservedwithbothcycloheximideandDOG.Theseresultssuggestthatpalatalepithelialcelldeathisnotapassiveevent,butanactiveprocessrequiringtheprogrammedsynthesisofspecificproteins.
