The consequences of X-dosage deficiency in the germ line: Impaired development in vitro of preimplantation embryos from XO mice☆

[摘要] Two-cellembryosfromXOandXXmicewereculturedfor3–4days.Thestageofdevelopmentreachedbytheembryoswasscoredatintervals,usingascriteriathenumberofcells,thedegreeofcompaction,andthepresenceorabsenceofablastocoele.TheresultsshowthattheembryosfromXOmicecleave,compact,andformblastocoeleslaterthandotheembryosfromXXmice.Furthermore,manyoftheXO-derivedembryosfailedtoformblastocystsandbecamegrosslyabnormal.ItissuggestedthatthisoverallimpaireddevelopmentisaconsequenceofX-dosagedeficiencyinthematernalgermlinethroughtheproductionofeggswhicharedeficientinX-linkedproducts.Inadditiontothisoveralldevelopmentalimpairment,afewembryosdiedattheeight-cellstage.ThesemayhavebeenYOembryos.ThebreedingcharacteristicsofXOmice,XX↔XYfemalemousechimeras,andthesterilityofXO(Turner'ssyndrome)womenarediscussedinthelightoftheseobservations.
