Regulation of lactate dehydrogenase (LDH) and alcohol dehydrogenase (ADH) synthesis in liver nuclei, following their transfer into oocytes☆
[摘要] TheregulatoryeffectofoocytecytoplasmonthesyntheticactivityoftransferredsomaticcellnucleiwasstudiedusinganinterspecifichybridcombinationofAmbystomatexanumandAmbystomamexicanum(axolotl).Theenzymeslactatedehydrogenase(LDH)andalcoholdehydrogenase(ADH)wereusedasmarkersofgeneactivity.Inbothspeciesofsalamanders,LDHissynthesizedintheliverandoocytes,whileADHistissue-specificbeingsynthesizedintheliverbutnotoocytes.BothLDHandADHshowspecies-specificpatternsonstarchgelswhichpermitdetectionofenzymessynthesizedbytexanumlivernucleifollowingtheirtransferintoaxolotloocytes.Analysisofrecipientoocytesafter1–3weeksinculturerevealsthepresenceofnewlysynthesizedtexanumLDHbutnotADH.Theseresultsindicatethatthetransferredtexanumlivercellnucleicontinuetosynthesizeaproduct(LDH)foundinbothlivercellsandoocytes,butfailtosynthesizetheliver-specificproduct(ADH)whichisnormallyabsentinoocytes.Thus,inthecaseofADHandLDHtheoocytecytoplasmappearstobeabletoregulatethesyntheticactivityofthetransferredsomaticcellnucleisoastoconformtotheoocytes'normalsyntheticoutput.
