Study of differential collagen synthesis during development of the chick embryo by immunofluorescence: I. Preparation of collagen type I and type II specific antibodies and their application to early stages of the chick embryo☆
[摘要] Theaimofthisworkwastopreparespecificantibodiesagainstskinandbonecollagen(typeI)andcartilagecollagen(typeII)forthestudyofdifferentialcollagensynthesisduringdevelopmentofthechickembryobyimmunofluorescence.AntibodiesagainstnativetypeIcollagenfromchickcranialbone,andnativepepsin-extractedtypeIIcollagenfromchicksternalcartilagewereraisedinrabbits,rats,andguineapigs.Theantibodies,purifiedbycross-absorptionontheheterologouscollagentype,followedbyabsorptionandelutionfromthehomologouscollagentype,werespecificaccordingtopassivehemagglutinationtestsandindirectimmunofluorescencestainingofchickboneandcartilagetissues.AntibodiesspecifictotypeIcollagenlabeledbonetrabeculaefromtibiaandperichondriumfromsternalcartilage.AntibodiesspecifictotypeIIcollagenstainedchondrocytesofsternalandepiphysealcartilage,whereasfluorescencewithintercellularcartilagecollagenwasobtainedonlyaftertreatmentwithhyaluronidase.ApplyingtypeIIcollagenantibodiestosectionsofchickembryos,theearliestcartilagecollagenfoundwasinthenotochord,atstage15,followedbyvertebralcollagensecretedbysclerotomecellsadjacenttothenotochordfromstage25onwards.TypeIcollagenwasfoundinthedermatomalmyotomalplateandpresumptivedermisatstage17,inlimbmesenchymeatstage24,andintheperichondriumoftibiaeatstage31.
