Thyroid-hormone-induced differentiation and development of anuran tadpole hind limbs: Detection and quantitation of M-line protein and α-actinin synthesis☆
[摘要] Administrationoftriiodothyronine(T3)toStageVIIRanacatesbeianatadpolesinducesprecocioushindlimbdevelopment.Musclefiberformationinundifferentiatedhindlimbtissueisfirstevidentbyphasemicroscopyinthethighregion48hrafterT3treatment.Indirectimmunofluorescencestudies,usingantibodiestofrogskeletalmuscleM-lineproteinandα-actinin,demonstratethattheseproteinsareinitiallydetectableinthethighregionwithin24hrafterhormoneadministration.Withincreasingtimeafterhormonetreatment,myofibrillarproteinandmusclefiberformationareenhancedinthethighregionandprogressfromthethightothedistalareasofthelimb.Quantitativeimmunochemicaldeterminationsofthesynthesisofα-actininandM-lineproteinindicatethattheirratesofsynthesisincreasemarkedlyandsynchronouslyinthe24hrfollowinghormonetreatment.Thesequantitativedata,inconjunctionwiththeimmunofluorescenceandcytologicalresults,provideevidenceforthyroidhormoneinvolvementinamphibianlimbmuscledifferentiationanddevelopment.
