Promotion of retinal neurite outgrowth by substratum-bound fibronectin☆
[摘要] Wehaveexaminedconditionsunderwhichaggregatesofembryonicchickneuralretinawillextendneuritiesinvitro.Trypsin-dispersedcellsfrom7-dayembryonicchickneuralretinawereaggregatedinrotationculturefor8hrandmaintainedinserum-freemediumonavarietyofstandardculturesubstrate.Aggregatesextendfewneuritesonuntreatedplastic,glass,orcollagensubstrata.However,pretreatmentofthesesubstratawithhumanplasmafibronectinenhancestheircapacitytosupportretinalneuriteoutgrowth.Aggregatesculturedonfibronectin-treatedsubstrataextendlong,radiallyorientedneuriteswithin36hrinvitro.Themorphologyoftheseneuritesisdistinctfromthatseenwhenaggregatesareculturedonpolylysine-treatedsubstrata.Inthelattercase,neuritesarehighlybranchedandgrowconcentricallyaroundtheaggregateperimeter.Additionoffibronectintopolylysine-treatedsubstratastimulatesradialneuriteoutgrowth.PromotionofneuriteoutgrowthisdependentontheamountoffibronectinboundtotheculturesubstratumandonthepHatwhichbindingoccurs.Therequirementsforfibronectin-mediatedneuriteoutgrowtharemorestringentthanthosepreviouslyreportedforfibroblastattachmentandspreading.
