Isolation of cloned genes differentially expressed at early and late stages of Drosophila embryonic development
[摘要] WehavepreparedcDNAlibrariesfrom2-to5-hrDrosophilaembryosorfromembryoswiththedorsalphenotype.BoththecDNAlibrariesandagenomiclibrarywerescreenedbydifferentialhybridizationtopoly(A)+RNAsequencesfromdifferentembryonicstagestodetectnewsequencesspecificallyappearinginthecytoplasmicpoly(A)+RNAat2–5hrorinthelatedorsalembryo.Thedevelopmentalprofileoftheclonesisolatedconfirmedthatsomeofthesesequencesarespecificallyfoundinearlyembryogenesisandmightperhapsbeinvolvedinearlydeterminationevents.Someofthelatedorsalsequencesappearverylateinthedorsalembryosandarenotdetectableinwild-typeembryos,suggestingthattheymightbespecificforlarvalstages.Amongourclonesaresomecontainingsequencesofdispersegenefamilies.Poly(A)+RNAofanewgenefamily,B104,beginstoaccumulateat2–5hr.ThecopiaRNAsequencebecomesabundantfrom10hron,whilethe412sequenceisdetectableonlyatlatestages(15–20hr).OurresultsindicatethattheappearanceofRNAsofavarietyofgenes,includinggenefamilies,isvariouslyregulatedduringdevelopment,possiblyattheleveloftranscriptionorpolyadenylation.
