Cell lineage analysis of cerebellar Purkinje cells in mouse chimeras☆
[摘要] Murinechimerasprovideanexperimentalsysteminwhichcelllineageanalysisofthemammaliancentralnervoussystem(CNS)canbeaccomplished.UtilizingacellmarkersystemthatpermitstheidentificationofcellsofeachgenotypeinvariouscellpopulationspresentinhistologicsectionsoftheCNSatdifferentdevelopmentalperiods,fatemapsofthemammalianCNScanbeconstructed.Thus,thepresenceorpersistenceofclonesofcellscanbereadilyvisualizedinsimplyorganizedCNSregions,likethecerebellarcortex.Theelectrophoreticvariantsoftheglycolyticenzyme,glucosephosphateisomerase(GPI,E.C.5.3.1.9;GPI-1A,GPI-1B),arethegenotype-specificcellmarkersmostcommonlyusedbyexperimentalmammalianembryologistsinstudiesofcelllineageutilizingmammalianchimeras.WehaveadaptedthiscellmarkersystemtopermitthevisualizationandunequivocalidentificationofcellscontainingtheGPI-1BvariantthroughouttheCNSofadultBALBcByJa3C57BL6Jchimericmice.Utilizingallozyme-specificanti-GPI-1Bantiserainimmunocytochemical(PAP)stainingtechniques,wecanscoresmallaswellaslargecellpopulations,neuronsaswellasglia.WehavereconstructedandstatisticallyanalyzedthelocationanddistributionofchimerismpresentinthePurkinjecellpopulationoffourofthesechimericmice.WefoundthePurkinjecellsineachoftheseanimalsexistedassmall(3–8)cellpatchesoflikegenotypethatwerenotrandomlyarranged.Thissuggeststhatclonesofcellsmaypersistascontiguousgroupsofcellsthroughoutmammaliancerebellardevelopment.
