Translational control in sea urchin eggs and embryos: Initiation is rate limiting in blastula stage embryos☆
[摘要] Todeterminewhetherinitiationisrate-limitinginproteinsynthesisduringtheembryogenesisofseaurchins,polyribosomeprofilesofunfertilizedeggsandcleavage,blastulaandprismstageembryoswereexaminedafterincubationoftheeggsandembryosinthepresenceandabsenceoflowamountsofemetine,aninhibitorofpolypeptideelongation.Theribosomeswereradioactivelylabeledwith[3H]uridinebyinjectionoftheadultsduringoogenesissothatwecouldmonitoremetine-dependentshiftsofmonoribosomestopolyribosomes.Althoughinitiationisnotratelimitinginunfertilizedeggsor2-to16-cellembryosofStrongylocentrotuspurpuratus,itisratelimitinginblastulaandprismembryos.WesuggestthatinitiationbecomesratelimitingtoallowtheselectivetranslationofcertainclassesofmRNAduringlaterdevelopment.
