DNA synthesis during the change to pupal commitment of Manduca epidermis☆
[摘要] CytophotometricandcellsorteranalysisofFeulgen-stainednucleiofthedorsalabdominalepidermisofthetobaccohornwormManducasextathroughthefinallarvalinstarshowedthatboth2Cand4Cpopulationswerepresent.Atransientpopulationofoctaploidcells(30%)wasseenonDay3atthetimeoftheecdysteroid-directedreprogrammingofthecellsformetamorphosis.Thisspecificreplicationwasfoundtooccurinvitro,irrespectiveofwhetherthecellswereincubatedin20-hydroxyecdysonetoinducethechangetopupalcommitmentorinhormone-freemediaduringwhichtheyretaintheircommitmenttomakelarvalcuticle.Cytosinearabinoside(1and10μg/ml)inhibitedcellcyclingduringthetimeofthechangeofcommitmentbutdidnotpreventtheecdysteroid-directedchange.Thus,DNAsynthesisiscoincidentwith,butnotnecessaryfor,thegeneticreprogrammingofManducalarvalepidermis.
