Cell movement in somite formation and development in the chick: Inhibition of segmentation☆
[摘要] Thecontributionofactivecellmovementtosomiteformation(segmentation)andthelaterdispersalofthesomitesclerotomewasexaminedusingcytochalasinD(CD).Stage14–16chickembryosweregrownoverliquidmedium.After8hrinculture,controlembryoshadanaverageofsixadditionalpairsofsomiteswhileCD(1–2μg/mldissolvedinDMSO)-treatedembryoshadnonewsomites.DMSOalonehadnoeffectonsomitogenesis.CD-treatedembryostransferredtodrug-freemediumrecoveredandsegmentationresumed.NormalandCD-treatedsegmentalplateswereexaminedbySEM.Drug-treatedsegmentalplatecellsroundedup,consistentwiththeinteractionofCDoncontractilemicrofilaments.Embryoscultured8hrwithorwithoutCDwerefracturedthroughsomitepair20andexaminedbySEM.Inuntreatedembryosthesclerotomehaddispersedandwasmigratingtowardthenotochord.CDstoppedsclerotomedispersal.TotestwhetherCDinterferedwithelaborationofextracellularmatrixmaterialassociatedwithsomitedevelopment,incorporationof[3H]glucosamineandNa235SO4bysomitesandsegmentalplatewasdetermined.Therewasnodifferenceintotallabelincorporation.Molecular-weightprofilesofproteoglycanobtainedusingcontrolled-poreglass-beadcolumnsshowedonlysmallproteoglycansforbothtreatedandcontroltissues.Therefore,thealterationofsegmentationandsomitemorphogenesisbyCDwasnotduetodetectablechangesinproteoglycansynthesis.
