Differential expression of aortic and lung elastin genes during chick embryogenesis☆
[摘要] Theratiosoftropoelastinbtoaweremeasuredinchickaortaandlungduringembryogenesis.Theratesoftropoelastinaandbsynthesisweredeterminedinshort-termorganculture.Theresultsdemonstratedthatinlungtissuetheratioofthetwotropoelastinsremainedessentiallyconstant.Eachofthetropoelastinscomprised50%ofthetotalelastinsynthesis.Intheaortictissue,tropoelastinbrepresented70%ofthetotalelastininthe11-to13-dayembryosandincreasedto91%byDay16.TheseobservationsseenintheorganculturesystemwereparalleledinmeasurementsoffunctionalmRNAscodingforthetwoproteins.MeasurementsoffunctionaltropoelastinmRNAsfrombothlungandaortictissueswereperformedinamRNA-dependentrabbitreticulocytelysatesystem.AlthoughthechangesintheabundanceofthetropoelastinmRNAsrevealedthesametrendasthatseenintheorganculturedata,themagnitudeofthetropoelastinbtoaratiointheaorticorganculturewastwicethatdeterminedinthecell-freetranslationofaorticmRNAs.Thedataobtainedfrombothcell-freetranslationsandorgancultureexperimentsdemonstratethatthereisadifferentialexpressionofelastingenesduringaortadevelopmentwhichissignificantlydifferentfromthatfoundindevelopinglung.
